State Route 160 (SR160) is and east-west route beginning at the interchange with the Hume Freeway at Glenrowan to the small town of Running Creek on the Kiewa Valley Highway. The routes traverses through the wine growing region between Glenrowan and the Ovens Rivers, before crossing the Great Dividing Range to reach the Kiewa River. SR160 was an alternative route to the Ovens Highway (now Great Alpine Road) as it ran parallel on the southern side of the Ovens River between Wangaratta and Gapsted.

SR160 shared the Ovens Highway with SR156 between Gapsted and Ovens.

The route is entirely single carriageway, with occasional line marking on shoulders.

In 1996, SR160 was replaced by three routes as part of the Statewide Route Numbering Scheme:
- C522 along Glenrowan - Myrtleford Road;
- B500 along Great Alpine Road;
- C543 along both Happy Valley Road and Running Creek Road.
